The retina is a multilayered structure lining the posterior segment of the eye. It consists of photoreceptors and retinal pigment epithelial cells (RPEpiCs). RPEpiCs are located between the neurosensory retina and the choroid, forming the outer blood-retinal barrier that controls the chemical composition of the subretinal space. RPEpiCs exhibit a specific polarized protein distribution within the cell. In contrast to other epithelia that usually face an apical lumen devoid of matrix, the apical plasma membrane of RPEpiCs is in direct contact with the extracellular matrix. RPEpiCs play a key role in regulating the turnover of adjacent photoreceptor membranes, retinoid metabolism and protecting the retina from light-induced damage via intracellular melanin, suggesting that RPEpiCs may contribute to the pathogenesis of various ocular diseases such as proliferative retinopathy and age-related macular degeneration.

Immortalized Mouse Retinal Pigment Epithelial Cells were transduced with lentivirus carrying the SV40 gene and puromycin resistance. Tests confirmed that the cells are free from bacterial and fungal contamination, exhibit excellent proliferation potential, and can be passaged 10-20 times in vitro.
