DAG NOMAD Muscarinic M5 Receptor (M5R) cell lines stably co-express tag-free GPCRs and diacylglycerol (DAG) NOMAD Biosensor to monitor G-protein modulation by DAG release. DAG NOMAD Muscarinic M5 Receptor (M5R) Cell Line contains U2OS cells that stably express the following two constructs:

  1. human Muscarinic M5 Receptor (M5R) with no tag;
  2. DAG NOMAD Biosensor (Red Fluorescence).

DAG NOMAD Muscarinic M5 Receptor Cell Line allows to assay compounds analyzing the G-Protein signalling pathway by Gq/G11 involving receptor activation. When an agonist binds to M5 Receptor, the Gq/G11 protein is activated which, in turn, triggers a cellular response mediated by DAG via Phospholipase C stimulation. This leads to an increase of red fluorescence intensity of DAG biosensor. So, the activity can be easily quantified on living cells in a dose-response manner by fluorescence emission in a microplate reader.
