Description

Endothelin receptor type B (EDNRB) as a GPCR activates a phosphatidylinositol-calcium second messenger system. This EDNRB multiplexed assay cell line assay compounds or analyze their capability to modulate EDNRB. When a ligand binds to the EDNRB, it activates a G protein, which in turn, triggers a cellular response comprising both Calcium and cAMP. This cell line allows measuring cAMP and calcium increase in the cytosol and analyzing the intensity and distribution of cAMP and calcium biosensors within the cell. This highly reproducible assay has been validated using Endothelin as agonist and measuring by both fluorescence intensity and image analysis. Each vial of EDNRB multiplexed assay cells stably express cAMP Nomad-FP650 biosensor, Calcium Nomad-tGFP biosensor and Endothelin receptor B (with no tag).
